Peptide Name: ZAK (158-165) pT161+pT162
Product Use: Services as a blocking peptide for use with the MLTK-pT161+pT162 rabbit polyclonal antibody (Cat. No.: AB-PK719) that is also available from Kinexus. This phosphopeptide may also be useful as a substrate for screening the phosphatase activity of protein phosphatases. The peptide sequence is located in the protein kinase catalytic domain activation T-loop between subdomains VII and VIII. T161 and T162 phosphorylation stimulates phosphotransferase activity.
Peptide Production Method: Solid-phase peptide synthesis
Peptide Origin: Homo sapiens
Peptide Sequence: HNH-pT-pT-HMS
Peptide Modifications N Terminus: Free amino
Peptide Modifications C Terminus: βAla-Cys
Scientific Background: MLTK (ZAK) is a protein-serine/threonine kinase of the TKL group and MLK family. It is activated by phosphorylation by PKN1 and autophosphorylation on T161 and S165. It has a pro-apoptotic activity and mediates cell cycle arrest and checkpoint regulation. It regulates the JNK and p38 pathways. Involved in regulation of S and G2 cell cycle checkpoints by direct phosphorylation of Chk2. MLTK causes cell shrinkage and disruption of actin stress fibers and phosphorylates histone H3 at S28. It has been suggested to play a role in cell transformation and oncogenesis.
