Scientific Background: PRKD2 (PKD2) is a protein-serine/threonine kinase of the CAMK group and PKD family. It lengthens the signal induced by diacylglycerol (DAG) by modulating the cascade downstream from PKC. It is activated by DAG and phorbol esters. Phorbol-ester/DAG-type domains bind DAG, mediating PRKD2 translocation to membranes. Autophosphorylation of S710 and phosphorylation of S706 by PKC relieves auto-inhibition by the PH domain PRKD2 can mediate NF-kB activation (induced from oxidative stress), cell proliferation (induced from MAPK1/3 signalling), inhibition of the transcription repression for HDAC7, T-cell antigen receptor signalling cascade, golgi apparatus vesicle trafficking, cell adhesion, angiogenesis, and secretory granule release.
