Scientific Background: LOK (STK10) is a protein-serine/threonine kinase of the STE group and STE20 family. It can control lymphocyte migration via its phosphotransferase activity. LOK has been shown to phosphorylate MSN and has been implicated in phosphorylating Plk1. LOK can negatively regulate MEKK1/MAP3K1, and may also mediate cell cycle progression. A K65I mutation in LOK seen with Testicular germ cell tumour (TGCT) can induce loss of kinase phosphotransferase activity.
