Product Use: Services as a blocking peptide for use with the BMPR2-pS375 rabbit polyclonal antibody (Cat. No.: AB-PK544) that is also available from Kinexus. This phosphopeptide may also be useful as a substrate for screening the phosphatase activity of protein phosphatases. The peptide sequence is located in the protein kinase catalytic domain activation T loop region between subdomains VII and VIII. S375 phosphorylation is predicted to be stimulatory for phosphotransferase activity.

Scientific Background: BMPR2 (bone morphogenetic protein receptor type II) is a protein-serine/threonine kinase of the TKL group and STKR family. It is a transmembrane receptor kinase that is a member of the bone morphogenetic protein (BMP) receptor family. BMPR2 functions to bind BMPs, which are named for their role in endochondral bone formation and embryogenesis. Activated by binding bone morphogenetic protein. This type II receptor phosphorylates and activates type I receptors which autophosphorylate, then phosphorylate SMAD transcriptional regulators. Insertional mutagenesis studies in mice support a role for this protein kinase in mouse cancer oncogenesis.
