Scientific Background: Pyk2 (PTK2B, FAK2) is a protein-tyrosine kinase of the TK group and Fak family. It is a non-receptor kinase that has a role in actin cytoskeleton rearrangement, cell migration, cell polarization, adhesion, bone remodeling, and controlling amount of marginal B-cells' migration and presence in the spleen. Phosphorylation at TY402 increases phosphotransferase activity and induces interaction with ErbB2, RasGAP and Src. Phosphorylation at Y579 increases its phosphotransferase activity. NPHP1 interaction with Pyk2 has been interrupted with a P859A mutation. A phosphorylation site of Pyk2 can be inhibited with an Y881F mutation, which will also impair GRB2 interaction.
