Scientific Background: PKCi (PRKCI, PKC-iota) is a member of the AGC group of protein-serine/threonine kinases in the PKC family, and Iota subfamily. This kinase is highly expressed and widely distributed in most tested human tissues. Phosphorylation of Y325, T403 and possible T412 of PKCi increases phosphotransferase activity. PKCi has been linked with the development of melanomas (metastatic). The amino acid sequence of PKCi showed greatest homology to PKCzeta, with 72% identity overall rising to 84% in the catalytic domain. PKCi has been implicated in Ras signalling and is a critical downstream effector of oncogenic Ras in the colonic epithelium. Transgenic mice expressing constitutively active PKCi in the colon are highly susceptible to carcinogen-induced colon carcinogenesis.
