Scientific Background: PDHK1 (PDK1, pyruvate dehydrogenase kinase 1) is a protein-serine/threonine kinase of the Atypical group and PDHK family. It functions in the regulation of glucose and fatty acid metabolism through the phosphorylation of the subunits PDHA1 and PDHA2 of pyruvate dehydrogenase. This inhibitory phosphorylation reduces metabolite flux into the trichloroacetic acid (TCA) cycle, decreases aerobic respiration, and prevents the formation of acetyl-CoA. In addition, PDHK1 functions to prevent apoptosis in cells exposed to hypoxia or oxidative stress. Due to the presence of HIF-1, PDHK1 has shown to have increased activity in hypoxic cancer cells. PDHK1 shunts pyruvate away from the citric acid cycle and helps to maintain hypoxic cell viability. PDHK1 may be a target for antitumour therapy, since PDK1 prevents apoptosis in these cancerous cells.
