Scientific Background: JNK1 (MAPK8) is a protein-serine/threonine kinase of the CMGC group and MAPK family. It is involved in the regulation of several cellular processes, including cell proliferation, differentiation, migration, transformation, and apoptosis, through the JNK/SAP signalling pathway. This pathway is activated by extracellular stimuli such as pro-inflammatory cytokines or physical stress, as well as signals of intracellular stress, such as misfolded proteins in the endoplasmic reticulum. Phosphorylation of T183 and Y185 by either of two dual specificity kinases, MAP2K4 and MAP2K7, increases phosphotransferase activity. It is inhibited by dual specificity phosphatases, such as DUSP1, which target the T183 and Y185 activatory phosphorylation sites.
