Scientific Background: GRK2 (also known as beta-adrenergic receptor kinase 1) is a member of the AGC group of protein kinases in the GRK family, and BARK subfamily. This kinase is highly expressed and widely distributed in most tested human tissues. Orthologues of GRK2 are highly conserved in vertebrates, including amphibians. It is activated by binding a Gbeta/gamma subunit complex. Phosphorylation of S29 increases its phosphotransferase activity. It specifically phosphorylates the agonist-occupied form of the beta-adrenergic and closely related receptors, probably inducing a desensitization of them. It is a key regulator of LPAR1 signalling, by competing with RALA for binding to LPAR1. It desensitizes LPAR1 and LPAR2 in a phosphorylation-independent manner.
