Scientific Background: BRSK1 is a protein-serine/threonine kinase of the CAMK group and CAMKL family. It is activated by phosphorylation on T205 by STK11 in complex with STE20-related adapter-alpha (STRAD alpha) pseudo kinase and CAB39. Its substrates include CDC25B, CDC25C, MAPT/TAU, RIMS1, TUBG1, TUBG2 and WEE1. Its phosphotransferase activity is lost with the K59A, and G327A mutations. It is involved in the UV-induced DNA damage checkpoint response, likely by inhibiting CDK1 activity through phosphorylation and activation of WEE1, and inhibition of CDC25B and CDC25C. A LKB1/LKB1 complex with the STE20-related adapter-alpha (STRADA) pseudo kinase and CAB39 phosphorylates and activates BRSK1, but this can be prevented by an inhibitory T189A mutation.
