Scientific Background: H-Ras is a small monomeric G protein that binds GDP/GTP and possess intrinsic GTPase activity. It alternates between an inactive form bound to GDP and an active form bound to GTP. It is normally activated by guanine nucleotide-exchange factors (GEFs) and inactivated by GTPase-activating proteins (GAPs). Mutations that compromise the GTPase activity of H-Ras (e.g. at G12) produce constitutively active, oncogenic forms of the protein, which commonly arise in human cancers.
