Scientific Background: PAK6 (PAK5) is a protein-serine/threonine kinase of the STE group and STE20 family. It can regulate gene transcription. MAPKK6/MAP2K6 or AR can induce PAK6 activity which will induce phosphorylation of the DNA-binding domain on an androgen receptor (AR) protein. The activation of PAK6 through MAP2K6/MAPKK6 can be inhibited through the three mutations S165A, S560A, and Y566F. It plays a role in the regulation of gene transcription. The kinase activity is induced by various effectors including AR or MAP2K6/MAPKK6. It phosphorylates the DNA-binding domain of androgen receptor/AR and inhibits AR-mediated transcription. It also inhibits ESR1-mediated transcription. It may play a role in cytoskeleton regulation by interacting with IQGAP1. It can protect cells from apoptosis through phosphorylation of BAD. PAK6 suppresses tumour progression through regulation of androgen receptor homeostasis.
