Scientific Background: MLK2 (MAP3K10) is a protein-serine/threonine kinase of the TKL group and MLK family. MLK2 is involved in the JNK signalling pathway leading to c-Jun activation. MAP3K10 will activate JNK by phosphorylation. JNK signalling is found to promote apoptosis, neurodegeneration, cell differentiation and inflammation. It is involved in nerve growth factor induced neuronal apoptosis. Homodimerization via the leucine zipper domains is required for autophosphorylation on serine and threonine residues within the activation loop, which facilitate enzyme activation. As MAP3K10 is an important mediator of the pro-apoptotic JNK signalling pathway it may have a role as a tumour supressor protein. Mis-sense mutations have been observed in metastatic melanomas but have not been characterized functionally. MLK2 promotes the proliferation and decreases the sensitivity of pancreatic cancer cells to gemcitabine.
