Scientific Background: Wnk3 (PRKWNK2) is a protein-serine/threonine kinase of the Other group and Wnk family. Activation requires autophosphorylation of S308. Phosphorylation of S304 also promotes increased activity. Interestingly, Wnk3 lacks the catalytic lysine in subdomain II that is believed to be critical for phoshotransferase activity, and instead has a conserved lysine in subdomain I. It functions as a positive regulator of the transcellular Ca2+ transport pathway, and it plays a role in the increase of cell survival in a caspase-3-dependent pathway.
