Antibody Immunogen Description: Corresponds to amino acid residues K667 to I673; Just before kinase catalytic domain. This is the major in vivo phosphorylation sites.
Production Method: The immunizing peptide was produced by solid phase synthesis on a multipep peptide synthesizer and purified by reverse-phase hplc chromatography. Purity was assessed by analytical hplc and the amino acid sequence confirmed by mass spectrometry analysis. This peptide was coupled to KLH prior to immunization into rabbits. New Zealand White rabbits were subcutaneously injected with KLH-coupled immunizing peptide every 4 weeks for 4 months. The sera from these animals was applied onto an agarose column to which the immunogen peptide was thio-linked. Antibody was eluted from the column with 0.1 M glycine, pH 2.5. Subsequently, the antibody solution was neutralized to pH 7.0 with saturated Tris.This antibody was also subject to negative purification over phosphotyrosine-agarose.

Related Product 1: BUB1B-pS670 blocking peptide Scientific Background: BUBR1 (BUB1B) is a protein-serine/threonine kinase of the Other group and BUB family. It is an essential component of the mitotic checkpoint and is required for normal mitosis. It prevents progression of anaphase until all the sister chomatids are connected to spindles by binding to CDC20 to anaphase promoting complex (APC)/cycolsome. It is activated by the kinetochore motor protein CENPE, which increases phosphotransferase activity, and it is also involved in the localization of CENPE. BUBR1 negatively regulates Plk1 phosphotransferase activity in interphase cells, and it suppresses centrosome amplification and triggers apoptosis in polyploid cells that exit aberrantly from mitotic arrest. A K795R mutation inhibits the phosphotransferase activity of BUBR1, but does not abolish its ability to inhibit APC. BUBR1 may be a tumour suppressor protein (TSP). An R36Q mutation is linked with premature chromatid separation trait (PCS), which is an autosomal dominant disease. An R550Q mutation is found in Mosaic variegated aneuploidy syndrome 1 (MVA1), which is a severe developmental disease with characterized mosaic aneuploidies.
