Scientific Background: JAK1 is a widely expressed non-receptor tyrosine-kinase involved in the interferon-alpha/beta and -gamma signal transduction pathways, and can bind directly to the Interleukin 2 receptor. It couples cytokine ligand binding to tyrosine phosphorylation of various known signaling proteins and of a unique family of transcription factors termed the signal transducers and activators of transcription, or STATs. It is known to interact with IL31RA, IFNAR2, JAKMIP1 (via FERM domain) and SHB. It is expressed at higher levels in primary colon tumors than in normal colon tissue, but its expression level in metastatic colon tumors is comparable to the level in normal colon tissue. It is also activated in B cell lines from patients with post-transplant lymphoproliferative disorder, and in a mouse Lck-driven T cell lymphoma. JAK family kinases feature two protein kinase domains. Piceatannol is a known JAK1 inhibitor.
