Production Method: The immunizing peptide was produced on an A.B.I. automated peptide synthesizer. The peptide was cleaved from the synthesis resin with HF and purified by reverse-phase hplc chromatography. Purity was assessed by analytical hplc and the amino acid sequence confirmed by amino acid analysis. This peptide was coupled to KLH prior to immunization into rabbits. New Zealand White rabbits were subcutaneously injected with KLH-coupled immunizing peptide every 4 weeks. The sera from these animals was applied onto an agarose column to which the immunogen peptide was thio-linked. Antibody was eluted from the column with 0.1 M glycine, pH 2.5. Subsequently, the antibody solution was neutralized to pH 7.0 with saturated Tris.

Scientific Background: CDK8 is a protein-serine/threonine kinase of the CMGC group and CDK4 family. It is a component of the Mediator complex, which is a coactivator involved in regulated gene transcription of nearly all RNA polymerase II-dependent genes. This complex functions as a bridge to convey information from gene-specific regulatory proteins to the basal RNA polymerase II transcription machinery. The Mediator is recruited to promoters by direct interactions with regulatory proteins and serves as a scaffold for the assembly of a functional preinitiation complex with RNA polymerase II and the general transcription factors. It phosphorylates the CTD (C-terminal domain) of the large subunit of RNA polymerase II (RNAp II) to inhibit the formation of a transcription initiation complex. It phosphorylates CCNH leading to down-regulation of the TFIIH complex and transcriptional repression. It is also recruited through interaction with MAML1 to hyperphosphorylate the intracellular domain of NOTCH, and promote its degradation.
